Description

Description: This assay employs a two-site sandwich ELISA to quantitate MGST3 in samples. An antibody specific for MGST3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MGST3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MGST3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MGST3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Microsomal glutathione S-transferase 3 is an enzyme which catalyzes the conjugation of leukotriene A4 and reduced glutathione to produce leukotriene C4. This enzyme also demonstrates glutathione-dependent peroxidase activity towards lipid hydroperoxides.
MGST3 shares 36% amino acid sequence identity with MGST2, 27% identity with LTC4S, 22% identity with MGST1, and 20% identity with FLAP. In in vitro assays, recombinant MGST3 showed glutathione-dependent peroxidase activity and LTC4S activity. Northern blot analysis detected a 0.6-kb MGAT3 transcript in a wide variety of human tissues, with the highest expression in heart, skeletal muscle, and adrenal cortex
